A comprehensive review of the oxidative cyclisation of 2’-hydroxychalcones to aurones and flavones

The oxidative cyclisation reactions of 2’-hydroxychalcones to corresponding aurones and flavones are discussed. While Hg2+, Cu2+ and Tl3+-mediated oxidative cyclisation of 2’-hydroxychalcones selectively give aurones, I2, Se4+, In3+, Cu+ and Fe3+-mediated reactions preferable give flavones. The general mechanisms of these oxidative cyclisation reactions are proposed.

Synthesis of cytotoxic ferrocenyl flavones via a ferricenium-mediated 1,6-oxidative cyclization.

The oxidation of the ferrocenyl group of 2'-hydroxyferrocenyl chalcones activates the beta-position of the unsaturated ketone to nucleophilic attack to yield the first examples of ferrocenyl flavones. These compounds are significantly more cytotoxic than their organic analogs on B16 melanoma cells, with IC(50) values in the low micromolar range.
